Exceprt from my /sys/endian.h, which came with DJGPP 2.1:

#define LITTLE_ENDIAN          1234
#define BYTE_ORDER             LITTLE_ENDIAN


Just #include endian.h and test with

#if BYTE_ORDER   = LITTLE_ENDIAN etc.
