(1) You shouldn't mess with BIOS data directly.
Use some higher level API instead.
(2) If you really have to, use:
unisgned char bytevalue; /* assuming it is a 8-bit value at 40:42 */
dosmemget (0x442, sizeof(unsigned char), &bytevalue);
dosmemput (&bytevalue, sizeof(unsigned char), 0x442);
